Combined Metabolome and Transcriptome Analysis of Floral Organ Development in Magnolia cavaleriei var. platypetala ‘Tanchun’

by Yingbing Hu, Zhe Zhang, Yubing Yong, Minhuan Zhang, Xijun Hu, Weiqun Lei, Donglin Zhang

Magnolia cavaleriei var. platypetala ‘Tanchun’ is a newly registered flower variety in China, known for its characteristic floral aroma that intensifies toward full bloom. However, the composition of the volatiles of this aromatic flower remains uncharacterized. Here, we compared the volatile organic compound composition of Tanchun through gas chromatography–mass spectrometry and comparative transcriptome sequencing analyses of the stamen (S), pistil (P), and petals (T) during flower development, i.e., the bud (S1), semi-opened (S2), and bloom (S3) stages. We present a first comprehensive profile of 1395 metabolites from Tanchun’s floral organs. Terpenoids (26.2%) constituted the largest chemical group, followed by esters (17.52%), nitrogen compounds (9.83%), hydrocarbons (8.11%), alcohols (7.97%), aldehydes (6.53%), and others. We found that volatile organic compound (VOC) accumulation was both spatiotemporal and stage-specific. The S1 and S2 transition was characterized by scent notes of green, herbal, and waxy aromas, while the S2 and S3 shift exhibited a richer profile of fruity, sweet, and creamy notes, primarily in petals. A comparative VOC and transcriptomic analysis revealed that petals activate pathways for structural expansion and precursor mobilization, stamens enhance lipid and terpenoid metabolism, and pistils maintain a conserved profile. Importantly, the S1 and S2 transition in petals establishes the biochemical foundation by activating acyl-CoA, phenylpropanoid, and terpenoid synthesis pathways, which enables the activation of the butanoate metabolism pathway at S3, leading to the production of ester-rich compounds that define the floral scent. The transition to full bloom involves a shift to energy-efficient volatile biosynthesis, supported by carbohydrate restructuring and phytohormonal regulation. Our results provide the first comprehensive volatilome and transcriptome resource for ‘Tanchun’, revealing a highly efficient, multi-stage strategy for floral fragrance biosynthesis. This work lays a molecular foundation for future horticultural improvement and biotechnological applications in the flavor and fragrance industries.

Flowering Dynamics, Pollen Viability and Stigma Receptivity of Nai Plum (Prunus salicina Lindl. var. cordata) from Different Provenances

by Juan Luo, Yao Li, Fengxia Shao, Sen Wang, Kuo Yang, Tian Xiang, Xuanyu Zhang, Yutong Li, Xinxin Lian, Minhuan Zhang, et al.

Nai plum (Prunus salicina Lindl. var. cordata) is a high-value fruit crop in southern China, yet its post-harvest quality is often compromised by fruit browning, a major constraint to storage and marketability. Addressing this challenge requires a deeper understanding of the species’ reproductive biology, which underpins both fruit set and cultivar improvement. In this study, we characterized the flowering biological characteristics of Nai plum accessions introduced from Yanling and Liuyang (Hunan Province) and Shaoguan and Lechang (Guangdong Province). Using field observations combined with microscopic and submicroscopic techniques, we documented flowering phenology, flowering dynamics, floral organ traits, pollen viability and stigma receptivity. The flowering period was in March, lasting 26–28 d, and the group blooming period was divided into three stages: Initial opening stage, Full blooming stage, and Final flowering stage. The single-flower opening process was divided into eight stages. Pollen viability followed a unimodal curve, peaking at the petal flattening stage (PF) across all accessions, though peak values varied by provenances. Stigmas were of the wet type, with receptivity following a weak–strong–weak pattern; peak receptivity occurred at early flowering (EF) and PF in most accessions. The EF of Nai plum from Yangling (S1) lasted for 7 h, and PF lasts for 28 h. The EF of Nai plum from Yangling (S2) lasted for 3 h, and the PF lasted for 11 h. Both the EF and the PF of Nai plum from Shaoguan (S3) lasted for 14 h. The bud white stage (BW) of Nai plum from Lechang (S4) lasted for 6 h and the EF lasted for 7 h. The EF of Nai plum from Liuyang (S5) lasts for 7 h, and the PF lasted for 28 h. These findings clarify the reproductive phenology and floral biology of Nai plum, providing foundational knowledge that can inform breeding strategies and cultivation practices aimed at improving fruit set and, ultimately, post-harvest quality.

Diversity Analysis of Leaf Phenotypic and Fruit Quality Traits Among Six Superior Trees of Nai Plum (Prunus salicina Lindl. var. cordata)

by Kuo Yang, Juan Luo, Fengxia Shao, Sen Wang, Yao Li, Tian Xiang, Xuanyu Zhang, Yutong Li, Xinxin Lian, Minhuan Zhang, et al.

This study analyzed the phenotypic and internal fruit quality diversity of six superior Nai plum trees to provide detailed phenotypic profiles and preliminary relational hypotheses, supporting superior genotype re-selection for breeding. Using leaves and mature fruits, we conducted diversity, correlation, and principal component analysis (PCA) on all quantitative traits. The average Shannon–Wiener index (H′) for qualitative traits was 0.543, and the average coefficient of variation for quantitative traits was 19.98%. Correlation analysis revealed complex trait relationships, including the synchronous variation between the total number of soluble solids (TSS) and reducing sugars (RS) or soluble sugars (SS) and the opposite trends between the TSS and potassium (K), magnesium (Mg), or soluble protein (SP). PCA extracted four principal components (cumulative contribution: 91.074%) from all traits. Based on factor scores, S6 ranked highest, indicating its potential as a comprehensive candidate. The findings offer a theoretical basis for Nai plum cultivation and breeding.

Systematic Analysis of Reproductive Barrier Types and Strengths in Interspecific Hybridization Involving Magnolia crassipes

by Zhe Zhang, Yingbing Hu, Chenfei Huang, Minhuan Zhang, Xingliang Wu, Xiaoling Jin, Yan Huang

Magnolia crassipes is a valuable species in Magnolia sect. Michelia known for its unique purple flowers, but interspecific reproductive barriers limit its use in breeding. Using M. crassipes as the maternal parent, we performed 13 pollination combinations (one selfed control and crosses with 12 taxa spanning five sections). We assessed reproductive processes from pollen–stigma interaction to seed and seedling performance, and verified hybrids using SSR markers. Reproductive barriers are strongly associated with phylogenetic distance, shifting from pollen-adhesion failure in crosses with donors from distant-section, to abnormal pollen-tube guidance in cross with M. denudata, and to fruit initiation in crosses with pollen donors from sect. Michelia. Among these Michelia-donor crosses, prezygotic barrier strength varied among combinations, as reflected by differences in stigma germination and ovule entry rates, which strongly influenced the potential for fruit set success. Postzygotic barriers further reduced reproductive success via seed abortion (peaking at 83.8%). However, all germinated hybrids exhibited normal early growth. Notably, backcrossing with the F1 hybrid M. ‘Danxia’ significantly improved reproductive compatibility (seed abortion rate 6.3% and germination rate 100%). This study clarifies the key barriers in M. crassipes hybridization and provides a basis and practical strategies for its genetic utilization.

Interaction of PsMYB4 with PsEGL3 inhibits anthocyanin biosynthesis in tree peony yellow flowers

by Xiaoning Luo, Sijie Huang, Fangjun Jiang, Qianqian Shi, Yafeng Wen, Mengchen Li, Minhuan Zhang, Yanlong Zhang

Tree peony is a traditional woody flower originating from China and possesses high ornamental value. The yellow cultivar is even more precious, known as the highest grade. However, the molecular mechanism underlying tree peony yellow flower formation is still unclear. Our present work identified two transcription factors (TFs), PsMYB4 and PsEGL3, which were highly expressed in yellow tree peony cultivar. Phylogenetic analysis indicates that PsMYB4 belonged to the R2R3-MYB repressor, while PsEGL3 was clustered into subgroup IIIf of bHLH family. Overexpression of PsMYB4 and PsEGL3 respectively in tobacco inhibited anthocyanin synthesis, with PsMYB4 overexpressing lines being more pronounced and the expression levels of structural genes NtC4H, NtCHS, NtCHI, and NtDFR were significantly downregulated, while the expression levels of structural genes in PsEGL3 transgenic lines showed no significant pattern. On the contrary, the expression of PsCHS, PsCHI, PsF3H and PsDFR increased in either PsMYB4 or PsEGL3 silencing tree peony petals, in which endogenous PsMYB4 and PsEGL3 genes were also inhibited to a certain extent. Yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays further confirmed PsMYB4 could interact with PsEGL3. Moreover, in dual-luciferase (LUC) assay, PsMYB4 and PsEGL3 synergistically suppressed the promoter activity of PsCHS, PsCHI, and PsDFR, thus inhibiting anthocyanin biosynthesis branch and leading to a metabolic flow towards the flavonol synthesis branch. These findings provide a new perspective for revealing the molecular mechanism of the yellow flower formation in tree peony.

Mining Heat-Resistant Key Genes of Peony Based on Weighted Gene Co-Expression Network Analysis

by Xingyu Yang, Yu Huang, Yiping Yao, Wenxuan Bu, Minhuan Zhang, Tangchun Zheng, Xiaoning Luo, Zheng Wang, Weiqun Lei, Jianing Tian, et al.

The RNA-Seq and gene expression data of mature leaves under high temperature stress of Paeonia suffruticosa ‘Hu Hong’ were used to explore the key genes of heat tolerance of peony. The weighted gene co-expression network analysis (WGCNA) method was used to construct the network, and the main modules and core genes of co-expression were screened according to the results of gene expression and module function enrichment analysis. According to the correlation of gene expression, the network was divided into 19 modules. By analyzing the expression patterns of each module gene, Blue, Salmon and Yellow were identified as the key modules of peony heat response related functions. GO and KEGG functional enrichment analysis was performed on the genes in the three modules and a network diagram was constructed. Based on this, two key genes PsWRKY53 (TRINITY_DN60998_c1_g2, TRINITY_DN71537_c0_g1) and PsHsfB2b (TRINITY_DN56794_c0_g1) were excavated, which may play a key role in the heat shock response of peony. The three co-expression modules and two key genes were helpful to further elucidate the heat resistance mechanism of P. suffruticosa ‘Hu Hong’.

Insight into the Molecular Mechanism of Flower Color Regulation in Rhododendron latoucheae Franch: A Multi-Omics Approach

by Peng Xiao, Hui Zhang, Qiulin Liao, Ninghua Zhu, Jiaao Chen, Lehan Ma, Minhuan Zhang, Shouyun Shen

Rhododendron latoucheae Franch. (R. latoucheae) is a valuable woody plant known for its high ornamental value. While purple flowers are a distinct and attractive variant phenotype of R. latoucheae, the underlying mechanism regulating its flower color is still poorly understood. To investigate the molecular regulatory mechanism responsible for the variation in flower color, we selected plants with white-pink and purple petals as the object and conducted analyses of metabolites, key genes, and transcription factors associated with flower color. A combined metabolome–transcriptome analysis was performed, and the expression of key genes was subsequently verified through qRT-PCR experiments. The results of our study demonstrated a significant enrichment of differential metabolites in the flavonoid metabolic pathway. Changes in anthocyanin content followed the same trend as the observed flower color variations, specifically showing significant correlations with the contents of malvidin-3-O-glucoside, dihydromyricetin, gallocatechin, and peonidin-3-O-glucoside. Furthermore, we identified three key structural genes (F3GT1, LAR, ANR) and four transcription factors (bHLH130, bHLH41, bHLH123, MYB4) that are potentially associated with the biosynthesis of flavonoid compounds, thereby influencing the appearance of purple flower color in R. latoucheae.

Effects of Three Exogenous Substances on Heat Tolerance of Peony Seedlings

by Jiaxin Guo, Yu Huang, Xingyu Yang, Wenxuan Bu, Jianing Tian, Minhuan Zhang, Kaili Huang, Xiaoning Luo, Ye Ye, Wen Xing, et al.

In this study, one-year-old seedlings of Paeonia ostii ‘Fengdan’ were used as materials. The method of spraying exogenous salicylic acid (SA), calcium chloride (CaCl2) and abscisic acid (ABA) was used. The effects of different concentrations of SA, CaCl2 and ABA on the heat tolerance of peony seedlings under high temperature stress were studied. The optimum concentration and mechanism of SA-, CaCl2- and ABA-induced heat tolerance of peony seedlings under high temperature stress were discussed. The results showed that 100 μmol/L SA, 40 mmol/L CaCl2 and 40 mg/L ABA had the best induction effect on the heat tolerance of peony. The effects of three exogenous substances on the heat resistance of peony seedlings were ranked as SA > CaCl2 > ABA. The three exogenous substances mainly alleviated high temperature damage by alleviating the degradation of chlorophyll (Chl), relative electrical conductivity (Rec) content, increasing superoxide dismutase (SOD) activity, soluble proteins (SPs), free proline (Pro) and soluble sugars (SSs) content, and reducing SSs content under high temperature stress. The five indices had the most significant effect on the three exogenous substances in improving the heat resistance of peony seedlings. These could be used as an index to identify the heat resistance.
